My work is focused on developing the ground vibrational states of S-adenosylmethionine (SAM) when bound to an enzyme lysine methyltransferase (MTs), SET7/9, as detectors of its solution state conformation, and as sensors of noncovalent interactions with SAM in the enzyme active site. In MTs active sites, alternate conformers of SAM can provide new structural leads to enhance drug-compound specificity, as different enzymes may possess unique alternate conformers. Frequencies observed in the spectrum from SAM when bound within active sites can be used as probes to detect any druggable alternate conformers and non-covalent interactions in MTs active sites, aiding in specificity improvement and understanding their impacts on catalysis. With over 5 years of experience in academia and industry, specializing in analytical method development, small and large molecule synthesis, purification, and characterization. Expertise in a wide range of analytical techniques such as HPLC/UPLC (IEX, SEC, RP, HILIC), LC-MS, NGC/FPLC, FTIR, NMR, Raman, SV-AUC, ELISA, AlphaLISA, etc. Demonstrated expertise in quality control, compliance, method, and process development, seeking to leverage my scientific knowledge and expertise in analytical chemistry to support clinical therapies.

Efficiently collaborated with computational laboratories to validate band assignments via Density Functional Theory (DFT) calculations and molecular dynamics.• Optimized synthesis and purification of S-adenosyl-L-methionine (SAM) for infrared spectroscopic studies. Engineered the expression and purification of Metk, an important enzyme in SAM synthesis, ensuring optimal enzymatic activity. Devised strategic employment of ion exchange and size-exclusion chromatography to overcome purification challenges, successfully separating SAM from excess sulfate salt and co-purifying counterions. Applied the optimized method to enzymatically synthesize and purify various isotopes of SAM, including D3-methyl-SAM, 13C-methyl-SAM, and 15N-SAM, leading to the acquisition of clean IR spectra. Produced the first-of-its-kind IR spectra for isotopically labeled SAM derivatives, thus propelling the project into its subsequent phase.• Mentoring, and training undergraduate students.Provided mentorship and training to more than 10 undergraduate students, resulting in their co-authorship in our publications and subsequent progression to graduate-level studies.
