Prominently delineating mechanism which leads to chronic and acute kidney disorder in animal models.     *My project at Feinberg School of Medicine revolves around pathogenesis of diabetic nephropathy. In the past six years, i got chance to delineate in detail molecular mechanism of acute and chronic kidney disease. * Methylation specific PCR indicated that our target enzyme (Myo-inositol oxygenase MIOX) is hypomethylated in hyperglycemic settings published in American journal of Pathology* Cisplatin is common anticancer drug, but is known for its nephrotoxicity. Using immunohistochemistry, florescence, Q-PCR, Methylation specific PCR, Immunoblotting, Cell culture, ELISA, Gel shift assay established that MIOX genetic deletion protect kidney against cisplatin induced injury (JASN, 2017)* Similarly, Tunicamycin ( inhibitor of Post translational modification) and long standing diabetes generate advance glycation end product showed accentuation of renal injury with MIOX overexpression ( AJP, 2018 & 2019). * Cell based assay and usage of various inhibitors suggested that MIOX overexpression exacerbates cell death in cisplatin induced AKI via accentuating ferroptosis and deletion attenuates renal injury ( JCI, 2019)* Used knockout and double knockout mice in diabetic and obese setting to establish the role of MIOX in diabetic nephropathy. * Present project: screening drug like small molecules which can potentially attenuate MIOX activity and contributes to attenuating progression of renal injury.* Skills developed and extensively used: Working with various disease mice models, Epigenetics, Cell culture, Generated overexpressing, knockout and mutated cell line, Gel shift assay, Subtractive hybridization, Histopathology, Immunohistochemistry, Cryosectioning, Immunofluorescence, Western blotting, ELISA, PCR, Q-PCR, In vitro transcription, Genotyping, Recombinant protein expression and purification, Cell based assay,Antibody generation, Random amplified cDNA ends, Co-culture ( cultured fibroblast and epithelial cell line).

My thesis revolved around elucidating the role of nuclear receptor peroxisome proliferators activated receptor-gamma (PPARg) in regulating steroidogenesis in ovary. * Using random amplified cDNA ends i was able to identify ovary specific novel isoform of PPARg which we named as PPARg1b (Gene, 2012)* We established serum free granulosa cell culture (DAE, 2011). * Elucidated the signaling mechanism regarding fertility regulation. Polyunsaturated fatty acid like conjugated linoleic acid (CLA) are administered in animals to treat negative energy balance. We established that CLA enhance ovulation via binding to PPARg and accentuating downstream signaling (Reproduction, 2012).
