Biophysics, Bioanalytics, Method development - Prof. Dirk-Peter HertenTitle: "Development of novel super-resolution and volume imaging techniques for visualising cellular function and ultrastructure in biomedical applications"The focus of this work was on the development of novel methods for the analysis of biomedically relevant structures, particularly through the establishment of quantitative techniques and correlative light-electron microscopy. Initially, I custom-designed and constructed a fully automated super-resolution fluorescence microscope to perform unsupervised sample acquisition. Samples then included primary cells such as liver sinusoidal endothelial cells. Here, the development of novel analytical approaches and the automation of processes to increase the accuracy of results were major focuses. Special emphasis was placed on the integration of state-of-the-art technologies and the programming of software-based analysis routines.1. Developing novel techniques in quantitative microscopy2.
