I joined the Weir Lab during the spring of 2018. During the summer of 2018, I was accepted to the summer research fellowship at Wesleyan University, and worked for 9 weeks as a wet lab researcher in the Weir Lab. During those nine weeks, Professor Weir and a graduate student, Will Barr, became my mentors, and I was able to work on my own independent research project. I learned many new procedures in the lab such as using TCA prep to isolate proteins from yeast cells, running Western-blots, and colony PCRs. Additionally, I attended weekly seminars and workshops, a symposium, various social events, and a closing poster session where I presented my research project. I also learned how to write my own scientific abstract. I received the same research fellowship (different source: CIS stipend) for the summer of 2019 and worked on a new independent project focused on optimizing a cycloheximide protocol for investigating the protein stability of genes of interest. I mentored a new undergraduate student in the lab, Jungwoo Cho, and worked together with her on the project.This work has been published in PLOS ONE. In the summer of 2020, I received the QAC Summer Apprenticeship stipend to continue doing research in the Weir Lab. Due to the coronavirus pandemic, I worked through remote research and focused on a different aspect of the project which was Molecular Dynamics (MD) simulations. I learned how to simulate the ribosome using MD and used the AMBER suite to analyze the data that was generated in the simulations. My independent project focused on changing the +1 codon just after the A-site codon and observing how the rRNA to mRNA interaction (through hydrogen-bonding) is affected.During the school semester, I continue to work in the wet/dry lab on various research projects.

I completed online laboratory safety trainings before closely working with my lab mentor, Dilduz Telman, and helped introduce several optimization changes to immune repertoire sequencing assay which aims to target and sequence heavy chains of T cells and B cells. On the job, I learned several important laboratory procedures such as isolating DNA from whole blood and PBMC samples, using Qubit 1x HS dsDNA to quantify DNA, and using the Agilent TapeStation to perform quality check and check sizing of DNA samples. I also learned how to operate NextSeq 550 to generate Next Generation Sequencing results from DNA libraries. Towards the end of the internship, I worked on an independent project to introduce the addition of primers to improve sensitivity of the immune repertoire sequencing assay. I also learned how to transfer raw NGS data into interpretable data by submitting it to bioinformatics pipeline. I would occasionally present lab results at meetings and created a final online research poster.
