    In Sue Biggins' lab, I developed an in vitro method to assemble the budding yeast kinetochore, a macromolecular protein structure. This assembly reaction appears to have the same fundamental requirements and characteristics as assembly of the kinetochore in vivo. It also generates functional kinetochores that are able to bind microtubules through the primary force-bearing interface of the kinetochore. I used this method as a tool to investigate the phosphoregulation of kinetochore assembly, directly demonstrate the order of inner kinetochore assembly, and reveal the differential roles of two separate pathways to outer kinetochore recruitment.
