Royal Microscopical Summer StudentRoyal Microscopical Society Jul 2021 - Oct 2021Nottingham, England, United KingdomI was awarded a grant from this society to do a research project in the University of Nottingham’s endothelial and vascular imaging lab with Dr Kenton Arkill (https://www.nottingham.ac.uk/medicine/people/kenton.arkill). At the end of this period, I produced a full report for publication in the Royal Microscopical Society’s ‘infocus’ magazine and a presentation which will be available for viewing on their youtube channel. The title of my project is ‘3D Nanoanalysis of the Tumour Microenvironment: Subcellular Ultrastructure of Model Systems’. In this project, I analysed HIF-1 and 2 knockout 3D spheroids, I looked at the changes in the spatial organisation and cell proportion in these spheroids as well as the orientation of the cells, by analysing the diameter of mitochondria in the different knockouts. I imaged these along the diameter in both low magnification and high magnification to look at the ultracellular structure. I used Fiji and Prism for the quantitative and qualitative analysis of these images.I have had full training on the Tecnai 12, a transmission electron microscope, and am able to use it confidently and independently. I also able to help with imaging on a scanning electron microscope in transmission mode and the staining and embedding of my spheroids.
