Center For Coastal Marine Sciences, Cal Poly, San Luis Obispo

In Dr. Alexis Pasulka’s Plankton Ecology and Coastal Oceanography Lab, I am involved in:Maintaining autotrophic and heterotrophic bacterioplankton cultures. I perform biweekly culture transfers using aseptic techniques while monitoring growth.Assisted in monthly water sampling at the Cal Poly Pier to characterize temporal variability of various microbial community characteristics in a coastal upwelling ecosystem. Lead CTD cast, collect and process water samples for DNA analysis. Characterization of harmful algal bloomer Akashiwo sanguinea through allelopathic/co-culturing experiments, transcriptome and bacterial analysis, growth analysis at varying temperatures, etc.Assisted in the optimization and testing of a quantitative PCR assay to screen for A. san's abundance in natural samples. This project involves the use of DNA extraction, gel electrophoresis, PCR, and quantitation methods for the detection of said organisms.

Indian Ocean

At-sea lead for Dr. Craig Carlson’s Lab at UCSB. Spent 55 days at sea aboard R/V Roger Revelle, independently sampling, preserving, and logging dissolved organic matter (DOM) samples. Specialized in the US GO-SHIP I05 line across the Indian Ocean from Fremantle, AUS to Cape Town, SA.

My NSF funded research in the Carlson Microbial Oceanography Lab at UCSB aims to understand the controls on the turnover of macroalgal dissolved organic carbon (DOC) in the ocean. I investigated the bioreactivity of compositionally-different Giant Kelp (Macrocystis pyrifera) DOC compounds and how the enzyme-inhibiting, M. pyrifera’s phlorotannins (polyphenols) regulate the turnover of DOC. This work explores the hypothesized controls on the bioavailability of DOC via heterotrophic bacterioplankton. By further examining controls on macroalgal DOC, we can better quantify kelp’s potential to be grown for atmospheric carbon dioxide removal.

Center For Applications In Biotechnology, Cal Poly, San Luis Obispo

Under the supervision of geneticist Dr. Jean Davidson, marine ecologist Dr. Crow White, and Masters Student Ben Daniels, I contributed to the identification of genes in Kellet’s whelk (Kelletia kelletii) populations that are adapted to local ocean conditions, such as temperature.Performed gDNA extractions from Kellet’s Whelk tissue using the salting-out protocol, with over 300 samples completed. I assist with managing tanks on the Cal Poly Pier. I read and discuss pertinent scientific articles in lab meetings.

Under the supervision of aquaculture specialist, Dr. Kevin Johnson, I contributed to the understanding of the genetic potential for adaptation to ocean acidification and hypoxia in the Pacific Oyster (Crassostrea gigas). I assisted with rearing oyster larvae, performing frequent counts of cultured larvae via microscopy, and oyster water tank changes at the Cal Poly Pier (Center for Coastal Marine Sciences).

Santa Cruz Island, California

As a field biology research assistant affiliated with the UC Reserve System and the Nature Conservancy, I worked closely with biologists and peers to contribute to ongoing botanical surveys on Santa Cruz Island (SCI). In preparation for the two research trips to SCI, I researched and prepared an oral presentation on the Island Fox's (Urocyon littoralis) decline and recovery effort to share with peers and biologists. I learned field identification skills of over 40+ plant specimens relating to the study. I was trained to utilize topographic maps, follow compass bearings, and use GPS devices. During the duration of the study, I surveyed insular vegetation utilizing several field techniques. I attended several presentations pertaining to ongoing research on SCI.I was honored with a position to lead a group of 15+ students on SCI. I was tasked with managing field data collected each day, organizing and filing paperwork, executing a plant identification test, maintinaing field equipment, and other general duties requested by field biologists.

As a field research assistant partnered with the LiMPETS student program and NOAA, I participated in ongoing studies pertaining to the health of California intertidal populations. Specifically, I contributed to the monitoring program for the Ochre Sea Star (Pisaster ochraceus) affected by the Sea Star Wasting disease. I used the transect and quadrat method at multiple tidepool sites to track Ochre Sea Star population density. I was trained on plant taxonomy, invertebrate zoology, and marine ecology. I analyzed and presented project findings to fellow peers and biologists.

Assisted undergraduate student and principal investigator, Dr. Emily Fogle, to determine the concentration of a specific protein, amylase, in a sample of biomolecules using the ELISA assay technique. I prepared different buffer solutions, prepared microtiter plates, ran ELISA assays, while taking precise notes and collecting data. Wrote written report summarizing efforts and presented findings to Dr. Fogle.
