In Dr. Alexis Pasulka’s Plankton Ecology and Coastal Oceanography Lab, I am involved in:Maintaining autotrophic and heterotrophic bacterioplankton cultures. I perform biweekly culture transfers using aseptic techniques while monitoring growth.Assisted in monthly water sampling at the Cal Poly Pier to characterize temporal variability of various microbial community characteristics in a coastal upwelling ecosystem. Lead CTD cast, collect and process water samples for DNA analysis. Characterization of harmful algal bloomer Akashiwo sanguinea through allelopathic/co-culturing experiments, transcriptome and bacterial analysis, growth analysis at varying temperatures, etc.Assisted in the optimization and testing of a quantitative PCR assay to screen for A. san's abundance in natural samples. This project involves the use of DNA extraction, gel electrophoresis, PCR, and quantitation methods for the detection of said organisms.

My NSF funded research in the Carlson Microbial Oceanography Lab at UCSB aims to understand the controls on the turnover of macroalgal dissolved organic carbon (DOC) in the ocean. I investigated the bioreactivity of compositionally-different Giant Kelp (Macrocystis pyrifera) DOC compounds and how the enzyme-inhibiting, M. pyrifera’s phlorotannins (polyphenols) regulate the turnover of DOC. This work explores the hypothesized controls on the bioavailability of DOC via heterotrophic bacterioplankton. By further examining controls on macroalgal DOC, we can better quantify kelp’s potential to be grown for atmospheric carbon dioxide removal.

Under the supervision of geneticist Dr. Jean Davidson, marine ecologist Dr. Crow White, and Masters Student Ben Daniels, I contributed to the identification of genes in Kellet’s whelk (Kelletia kelletii) populations that are adapted to local ocean conditions, such as temperature.Performed gDNA extractions from Kellet’s Whelk tissue using the salting-out protocol, with over 300 samples completed. I assist with managing tanks on the Cal Poly Pier. I read and discuss pertinent scientific articles in lab meetings.
