Design and synthesis of ring-fused Tricyclic 2-pyridone compounds with amyloid fibril binding activity

I worked on Rational Design and synthesis of Kinase Inhibitors for oncology drug discovery. Particularly my research work was focused on the heterocyclic scaffolds. I have synthesized two series of pyrimidine scaffolds and screened in cell based and enzymatic assays. The detailed mechanism of kinase inhibition was also studied. Molecular modeling studies were performed to demonstrate the type of inhibition, which was then validated with the biochemical assay.

Design and Synthesis of Pyrimidine Containing Heterocyclic Building Blocks

During the course of my dissertation work involved the development of metal-free and metal-mediated domino one pot synthesis of medicinally important heterocyclic scaffolds, and further their evaluation for potential biological activities. Our efforts led to development of 7 new synthetic methodologies for heterocycles such as pyrroles, imidazo[1,2-a]pyridines, imidazoles, bisindoles and quinolines. This work also involved development of a transition metal catalyzed C-H functionalization methodology. The molecular library prepared in each heterocylic scaffold was screened for P-gp modulatory activity to discover potential multi-drug reversal agents or amyloid-β clearing agents (anti-Alzheimer agents).
