Analysis of mutational signatures of low frequency variants in TRACERx primary and relapse samplesThe analysis of somatic mutation patterns has identified different mutational signatures that represent the activity of different exogenous or endogenous mutagenic processes that have been active during the development of tumours. Low-frequency variants are usually excluded from analysis because of their high potential to be artefactual. However, low-frequency mutations still contain a lot of information like potential mutagenic processes that are active subclonal as well as subclonal driver mutations. In this project, my role was to apply different de novo signature detection analysis tools to identify mutational signatures in low-frequency variants in TRACERx primary and relapse tumours. This was based on two different mathematical approaches, non-negative matrix factorisation (NMF) and hierarchical Dirichlet processes (HDP). In addition, my analysis exploits the differences between mutational signatures in primary versus relapse samples.
