Characterized endothelial cells (ECs) that were derived from induced pluripotent stem cells (iPSCs) of pulmonary arterial hypertension (PAH) patients. We showed that these cells had a similar phenotype to a patient's native endothelial cells, allowing for drug testing without the need for a lung transplant.Simulated the endothelial response to hemodynamic shear stress (blood flow) and characterized changes in the transcriptome, epigenome, and chromatin architecture. This identified mechanisms that underlie the inflammatory phenotype observed in atherosclerosis-prone regions of the vasculature.Identified a rare genetic variant associated with skeletal dysplasia and progressive joint stiffness.
