For 30+ years, I have been involved in microscopy-based research and tuition at Universities and Science Councils. The experienced I gained across a broad range of life and material science fields during that time has provided me with an enabling platform from which I can further impact scientific research and the development of much-needed microscopy skills. I have developed in-depth awareness of suitable sample preparation and imaging approaches if one is to get artefact- (deformation) free information from the samples of interest. While in most instances conventional preparation methods are suitable, in many others they are not. Using cryotechniques I have been able to access reliable information that wouldn't have been not possible with conventional (chemical) processing protocols. For example, in pulp and paper studies, I used freeze drying to visualise the development of microfibrils on wood fibres following beating, stages of cellulose degradation in dissolving pulp, and microtomy of embedded samples to study paper structure. In polymer nanocomposites studies, I routinely use cryosectioning not only to obtain sections for TEM but also cryo-planed SEM block faces to measure the thickness of layers in laminates, or to access the dispersion of nanocomposites, over much larger areas. Of course oil / water mixtures in foodstuffs and cosmetics are inaccessible by conventional imaging, as are aqueous fixation of partially hydrated living and non-living samples. And the list goes on (but I won't!)There is also a dire need to strengthen the microscopy skills of operators in many laboratories. In spite of the ever-increasing ease of use and capabilities of scanning and transmission electron microscopes, most operators - of course with notable exception - stick to the well-trodden routine operation without exploring the full capabilities of the instrument or its attachments, such as elemental microanalysis (EDS), cryo-transfer systems or tomography. The sample preparation caveat of "rubbish in, rubbish out" is often overlooked and evident in the outputs. This could be due either to staff turnover, or simply lack of applications training. Last is the often neglected quantification of information contained in images using freely available platforms such as ImageJ/Fiji. If it can be seen it can be measured, making results available for statistical analysis to attach a measure of confidence to the results reported. Senior Medical Natural Scientist, Electron Microscope Unit
I provided light and electron microscopy training and support to post-graduates and staff from a range of University Departments (Dental Health, Pharmacy, Biology, Chemistry and Physics), other Universities in South Africa (University of Pretoria, Witwatersrand, Stellenbosch University), Science Councils (CSIR) as well as Industry. I was an active participant in all the research projects undertaken, leading to numerous publications in rated Journals. I was also the sole operator of low temperature sample processing equipment and cryoSEM.I played a pivotal role in the application for funding to the National Research Foundation (NRF) for a F200 High Resolution TEM, as well as in designing the facilities to house it.

The National Centre for Nanostructured Materials at the CSIR is a Nanotechnology Innovation Centre established by the DST for the advancement of nanotechnology research in South Africa. I oversee the smooth operation of the extensive range of analytical equipment in place to support such research. Amongst the salient strengths of the Facility is the characterisation of polymer nanocomposites using cryoultramicrotomy in combination with high resolution TEM and AFM, SEM, LM as well as chemical characterisation using FTIR and Raman imaging.

The EM Unit provided analytical and imaging support to bio-medical and material sciences researchers at the University, as well as serving the needs of industry. I advised users on the best approach(es) to obtain answers to the questions sought, often becoming actively involved in many long term research projects. I managed the facilities at both Westville and Pietermaritzburg campuses, consisting of a wide array of light, electron beam and atomic force microscopes and ancillary sample preparation equipment with a strong bias towards cryo-techniques. One of my academic interests at the time was Cryobiology, specifically long-term preservation of desiccation-sensitive plant germplasm. Expertise in cryo-techniques was developed to gain insights into the state of cellular ultrastructure whilst in the frozen state, and to relate this to post-thaw survival in vitro.
Project Leader, Forestry And Forest Products Research Center
Pulp and paper research conducted at the Centre aimed at furthering understanding of the relationship between inter-fibre bonding and paper strength, as well as cell wall architecture and its impact on the preparation of highly-purified cellulose as a nano-technology substrate and raw material. Both electron beam and atomic force microscopies played an important role in gaining insights into fibre and cell wall ultrastructure.
