Assisted in the research and discovery efforts of peptide and small molecule therapeutics addressing unmet needs in a variety of fields (antibiotics, gastroenterology, and fertility). Synthesized analogs of pharmaceutically relevant compounds for downstream testing and characterization. Addressed adverse properties (proteolysis, poor solubility, poor permeability, etc..) by incorporation and/or removal of moieties with desirable/undesirable physicochemical and physiochemical properties. Synthetic routes were optimized for complex structures (multiple disulfide bonds, high molecular weight) to allow for generation of targets at an acceptable yield and purity. Auxiliary moieties were characterized to elucidate structure relationships and reduce experimental variables. New targets were synthesized and tested to determine project feasibility and proof of concept.

Project focused on the development of an in-house oligonucleotide synthesizer capable of generating pools of long oligonucleotide with high fidelity, large diversity at an ideal yield. Modeling, characterization and experiment design were carried out to identify critical parameters. Synthesizer was bench marked against a range of established synthesizers employing different approaches. Process optimization included optimization of light source, solid support, reagents, microfluidics, harvesting, purification and pool analysis. When applicable, test assemblies were created to isolate concerning factors and establish degree of impact on overall process.
