-Developed a deletion construct by knocking out an utert gene (telomerase) to study the haplo-insuffeciency of Ustilago Maydis.-Generated the utert U.Maydis complementation construct by cloning the carboxin resistence ORF into TOPO-UTERT-GFP vector at the SfiI restriction site. The vectors are linearized with SfiI and integrated in U.Maydis at the ip locus. Accomplished gene deletion and gene integration in U.Maydis through homologous recombination techniques. -Created utert deletion construct by using SfiI technique and screen the transformants with desired replacements with PCR. -Employed Echerichia coli strains DH5 and TOPO10 for all cloning applications. Used pCR2.1 TOPO TA for cloning of PCR products. Employed techniques like DNA isolation, Plasmid isolation, Ligation, RNA extraction, Bacterial Transformation, Bacterial Cell Culture for acquiring results.
