    I studied how DNA-cutting CRISPR proteins recognize and cut their target to enable more precise gene-editing tools and design proteins with novel functions. We described a conformational checkpoint that governs activation of Cas9 and identified a protein domain responsible for target sensing that was engineered to create a hyper-accurate Cas9 protein (HypaCas9). We also found that the Cas12 family of DNA-targeting CRISPR proteins have ideal properties for nucleic acid detection, which we developed into a programmable DNA detection technology called DETECTR. DETECTR accurately identified high risk HPV types in patient samples and functions as a platform for rapid and affordable diagnostics.

    To investigate renewable energy solutions, we engineered a "Bionic Leaf" that integrates a light-driven, water-splitting catalyst ("Artificial Leaf") with the hydrogen-metabolizing bacterium Ralstonia eutropha H16. We identified viable conditions for R. eutropha that mitigate production of reactive oxygen species by the inorganic catalysts. We also engineered R. eutropha to produce medium-chain fatty acids by expressing a thioesterase, and improved yields by identifying and knocking out metabolic sinks for fatty acids.

    The Culotta lab is interested in the role of metal ions and oxygen radicals in biology and disease. We found that nutrient depravation stress induced mitochondrial superoxide damage to iron-sulfur clusters in yeast central metabolism. This disruption resulted in a buildup of acetate that is exported to acidify the environment, which can improve yeast cell growth during glucose starvation.

    Understanding the molecular signals that trigger dormant or active tuberculosis (TB) disease is essential for drug discovery and public health efforts. Previous studies in the Steyn lab identified a putative sensor of oxidative stress, which may be responsible for the metabolic shutdown in dormant mycobacteria. We engineered a reporter system to evaluate conditions of oxidative stress in the TB analog, Mycobacterium smegmatis.
