I learn to use LC-MS/MS in diagnostics and analytics. In addition, I set up a protocol to detect several metabolites for which I need ion-pair liquid chromatography.

Used techniques:cell culture - western blot - immunoprecipitationProject:Deubiquitination can prevent degradation, thereby stabilizing it. I studied the interaction of a deubiquitinating enzyme with two substrates which are are targeted to the proteasomes by the same E3 ligase.

This assay was developed to detect acetyltransferase activity in mitochondria and consequently purify and find an mitochondrial acetyltransferase. An important aspect of the project became to troubleshoot the HPLC.

Keywords:Biochemistry - recombinant cloning Project:A4GALT is an enzyme that transfers galactose on top of lactosylceramide leading to globotriaosylceramide. In Fabry's disease, the enzyme removing the galactose is defective and since globotriaosylceramid did not seem to have an important function, the idea was to inhibit A4GALT to prevent accumulation of globotriaosylceramide. in this project I cloned A4GALT with a histidine or Myc tag for purificational reasons and I added a signal peptide from chitotriosidase for export outside the cell. After this recombinant cloning I did the first pilot experiments to express the clones in cells and to purify the A4GALT cloned proteins. Those proteins could be used in inhibition assays.
