This assay was developed to detect acetyltransferase activity in mitochondria and consequently purify and find an mitochondrial acetyltransferase. Keywords:Biochemistry - recombinant cloning Project:A4GALT is an enzyme that transfers galactose on top of lactosylceramide leading to globotriaosylceramide. In Fabry's disease, the enzyme removing the galactose is defective and since globotriaosylceramid did not seem to have an important function, the idea was to inhibit A4GALT to prevent accumulation of globotriaosylceramide. in this project I cloned A4GALT with a histidine or Myc tag for purificational reasons and I added a signal peptide from chitotriosidase for export outside the cell. After this recombinant cloning I did the first pilot experiments to express the clones in cells and to purify the A4GALT cloned proteins. Those proteins could be used in inhibition assays.
