In vitro readout monitored ATP activity (cell titer glo); while in vivo methods were measured by tumor regression. These results were added to technical report. BITE MOLECULE (IMMUNOTHERAPY) 2011-2012•Increased thru-put for flow cytometry primary T-cell assay by utilizing less toxic dyes that were read by separate lasers. Collaborated with colleagues and reviewed literature to create protocol with minimal supervision. •Investigated biodistribution of BiTE molecule in tumor-bearing, humanized, intra-peritoneal T-cell mouse model. T-cells were activated, expanded, and implanted in mice. BiTE was dosed intra-peritoneal, then tumor regression was measured and reported. •Looked to as a person to quickly understand new computer software and easily adopted techniques. Often tasked to train colleagues in innovative procedures with an eye on efficiency. •Generated IncuCyte assay for comparative response of normal and diseased fibrotic cells to apoptotic stimuli. Detected apoptotic differences by Western blot and HCS fluoroscopy. •Characterized receptor numbers on primary diseased cells and cell lines. Presented and discussed this data at working group meetings.•Continued lab management duties. -

Presented and discussed results with multiple departments at working group meetings. •Established tumor ex vivo protein extraction method for syngeneic and xenograft mouse models. Utilized this method to correlate protein levels to tumor regression after treatment with a macrophage antibody. Communicated methods to working group for the use in biomarker development and patient stratification. •Investigated differences in tumor regression results when model was moved to CRO. Microarray results revealed CRO was given a disparate cell line. •Used literature to develop state-of-the-art method of screening multiple cells lines for receptor of interest. •Collaborated with colleague to write study report on the effect of a macrophage antibody against an established tumor-bearing mouse model. •Evaluated biomarker activity in mouse toxicology model by screening serum for increased cytokine after administration of antagonistic antibody.
