Assessed the role of translesion synthesis (TLS) DNA damage tolerance pathway in tumorigenesis and cancer therapy.• Analyzed and compared the effect of TLS status on carcinogen, oncogene, and chemotherapy-induced tumorigenesis, mutational burden, and cosmic signature.• Performed a targeted CRISPR-Cas9 screen using custom sgRNA library, performed downstream bioinformatic analysis for hit identification and validated synthetic lethal interaction of TLS deficiency. Additionally, interrogated kinases and metabolites to understand their correlation with DNA damage response in absence of TLS.• Interrogated The Cancer Genome Atlas (TCGA) gene expression datasets and determined the extent to which TLS pathway gene expression levels are altered in representative cancers and whether expression levels of TLS pathway genes correlate with mutation load.• Published a first author and a co-author article. Two first author publications in pipeline.Identified targetable DNA damage dependencies of oncogene KRAS in pancreatic adenocarcinoma (PDAC).• Developed an isogenic cell line model system to study KRAS-induced replication stress in the preneoplastic stage of PDAC.• Carried out mRNA-seq and interrogated TCGA-PAAD database to identify DNA damage response genes overexpressed in PDAC.• Performed a targeted siRNA screen /CRISPR-Cas9 screen and identified DNA damage response genes that sustain KRAS-induced replication stress during preneoplasia.• Outlined the mechanism of TRIP13-mediated tolerance of KRAS-induced replication stress in PDAC.• One first author and two co-authors publications in pipeline.

• Developed bioassays for studying Topoisomerase-DNA covalent complexes, a biomarker for efficacy of topoisomerase targeting anticancer drugs.• Decoded biochemical and signaling pathways that affect repair of topoisomerase-DNA covalent complexes.
