Temporary position Duties and responsibilities :- Implementation of the untargeted metabolomics workflow for blood specimens from epidemiological studies. I have optimized the pre-existing workflow for untargeted metabolomics, by including automation of sample preparation and schemes for analysis of large-batches of samples. Robotic extraction of metabolites from blood samples using Perkin Elmer Janus liquid handling system. I have developed a range of robotic methods to perform aliquoting step as well as the extraction steps using a varitety of labwares including eppendorfs, 96-well plates, Agilent Captiva plates and glass vials. Around 3000 blood samples from a range of studies within exposomics and nutritech proejcts were prepared using these automated methods.Operation of Agilent UPLC-QTOF 6550 instrument :- It included routine preparation of the instrument by cleaning ion-source, tuning, calibrating, changing solvents and system suitability tests. Around 2000 samples were analyzed using a standarized LC-MS method. Raw data were processed by me using Agilent softwares Qual, DA Reprocessor and mass profiler professional for quality control checks and generation of peak-lists for each file. I am responsible for maintaining records for operation of the instrument, consumables and other biospecimens storage and handling. Construction of MSMS library for 635 pure standards: I took the initiative to developed a library of MSMS spectra using the standarized LC-MS method on the 6550 QTOF instrument. I acquired the pure standards on 96 well plates from IROA technologies, USA. A chart for analysis was prepared and data acquisition for these standard has been completed for reversed phase column.
