Sep 2016 - Feb 2020Frederick, MdProject 1) Utilizing an adoptive cell therapy approach in non human primates, our lab is studying factors that influence an effective antiviral T cell response during AIDS-virus infection. Through the engineered expression of cloned T cell receptors (TCR) and homing receptors, we are able to confer target specificity and directed localization to infused autologous T cells. Using this strategy, we aim to eliminate viral reservoirs in the lymph nodes of SIV infected rhesus macaques. More broadly, through this immunotherapy process, we are gaining translatable insights into the various qualitative factors that determine the efficacy and persistence of adoptively transferred cells. Project 2) In collaboration with the Viral Oncology Section at the LBR AIDS and Cancer Virus Program, we are also studying the highly varied human T cell responses to Kaposi's sarcoma-associated herpesvirus. Using an in vitro experimental approach, we are cloning virus-specific TCRs that are isolated from sero-positive individuals, and studying TCR clonotype avidity using functional immunoassays. From these studies, we aim to identify key T cell factors that determine an effective T cell response, and potential correlates of immune protection.
