• Used a non-human primate model (rhesus macaque) of early-life wildfire smoke exposure to understand the long-term effects of air pollution, focusing on fine particulate matter (PM2.5). • Effects of neonatal wildfire smoke exposure on the cardiac and pulmonary systems were investigated using high-resolution computed tomography (HRCT) in combination with simulated computational fluid dynamics and functional respiratory imaging technologies. Effects of early-life wildfire smoke exposure on the immune system were investigated using ELISA, multiplexed Luminex bioassays, and flow cytometry. Transgenerational effects were investigated by measuring the immune response and systemic inflammatory protein expression profiles of dams that were exposed as infants to wildfire smoke and their associated offspring that were not directly exposed to wildfire smoke. • Investigated the acute effects of the 2018 Paradise/Camp wildfire episodes on the expression of Pulmonary Surfactant Protein C (SP-C) in the alveoli of 1 y rhesus macaques that were exposed during early-life. Reported that animals that experienced early-life wildfire smoke exhibited reduced SP-C expression in alveolar type II epithelial cells (AEC II) via immunofluorescence microscopy, western blot, and real-time PCR. Reported that SP-C protein expression was dynamic in rhesus macaques under homeostatic conditions with the greatest expression occurring during early-life. • Investigated the effects of early-life environmental tobacco smoke (ETS) exposure on hematopoietic stem cell (HSC) differentiation in rodents. Develop a model of neonatal tobacco smoke exposure and monitored the potential for dendritic cell (DC) differentiation in bone marrow-derived HSC. Developed an adoptive bone marrow transfer model to determine the effects of early-life ETS exposure on altered myeloid activation and differentiation status.
• Identified novel microRNA (miR) that regulate effector cytotoxic T cell (CTL) differentiation. We determined that perforin was a major target of miR-139 by co-transducing CTLs with vectors expressing miR-139 and a GFP protein linked to the 3’UTR of perforin. A reduction in levels of GFP was observed in cells that expressed both vectors. Additionally, we used a perforin dependent listeria protection assay to determine if miR-139 had a negative effect on immunity. We observed a ten-fold reduction in the ability to clear listeria from the spleens of mice that were given CTLs overexpressing miR-139.• Small hairpin (sh) RNA screen for chromatin regulators that could affect the trans-differentiation of pre/pro-B cells into macrophages. We created a single cell clone of a murine transformed pre/pro-B cell line that expressed an inducible transcription factor. Upon induction of the inducible marker (human estrogen receptor), the recombinant transcription factor would translocate into the nucleus and induce macrophage specific gene expression. We optimized conditions for the screen and followed up by testing hits that could enhance the trans-differentiation process. Our hope was to uncover discrete populations that may arise during the conversion process by analyzing surface protein expression using mass cytometry.• Engineered reporter genes suitable for a multiplexed mass cytometry. Our goal was to create a multiplexed system that allowed for rapid identification of shRNA present in a large multi-parameter assay. This system would allow for competitive screening of a large number of hairpins present in an in vivo model.• Studied post-translational modifications on chromatin. We used site directed mutagenesis to create numerous expression vectors containing mutations in the catalytic and regulatory domains of a histone de-ubiquitinase complex.
