About Jeffrey Caplan
As the Director of Bioimaging, I have been committed to maintaining a shared user microscopy facility that emphasizes training the next generation of microscopists and collaborating with a wide range of scientists. The University of Delaware Bio-Imaging Center at the Delaware Biotechnology Institute currently has approximately 80 different research laboratories using our facility. We have six full time staff members to provide expert training and guidance on confocal, multiphoton, total internal reflection, wide-field fluorescence, super resolution, atomic force, laser capture microdissection and scanning and transmission electron microscopy. Although my background is in plant molecular and cell biology, I currently collaborate with researchers studying plants, animals, bacteria and yeast that work in fields ranging from material science to translational cancer research. Overall, my main goal is to continue the Bio-imaging center’s history of producing high quality data to complement, augment or elevate the science of our researchers.Specialties: I have a broad background in light microscopy, including experience with advanced light microscopy techniques, such as multi-photon, live-cell imaging, 4D acquisition, FRET, BiFC, FRAP, FLIP, photo-activation, total internal reflection microscopy, deconvolution, fluorescence anisotropy, fluorescence correlation spectroscopy, super-resolution and spectral unmixing. I believe that image analysis is a key aspect of modern microscopy and I have experience with a variety software packages, including ImageJ (Fiji), Imaris, Hyugens deconvolution, Amira and Volocity.

Associate Professor
CurrentAs faculty in the Department of Plant and Soil Sciences my research has two main focuses. The first is the role of organelle dynamics during plant innate immunity. Specifically, we are interested in how and why chloroplasts dramatically change their morphology by sending out stroma-filled tubules called “stromules.” We are studying their function during chloroplast movement and localized release of signals, such as reactive oxygen species (ROS), during innate immune responses to pathogens. The other focus of our lab is the development of novel microscopy methods using a variety of model systems, that includes bacteria, fungi, mammalian cell lines, and plants. Currently, we are creating methods for live-cell small RNA detection, quantitative super-resolution RNA imaging, deep learning image analysis, and plant tissue clearing.
