Led and implemented projects related to the RISC (RNA Induced Silencing Complex), the Argonaute-2 protein, and their interactions with a variety of partially Thiomorpholino-modified RNAs, in the pursuit of developing a therapeutically viable siRNA. Developed and optimized custom assays based on literature to determine AGO-2 cleavage efficiency. Worked with multiple labs to confirm and further explore exciting and irregular findings. Work required in-depth experimental planning, troubleshooting, usage of complex equipment, image analysis and interpretation, and tact when performing protocols with sensitive RNA compounds.Work resulted in the publication and defense of an Honors Thesis titled, "Exploring Partially Morpholino-Modified siRNAs and their Interactions with the Argonaute-2 Protein", as well as a research presentation given at a national conference (NCUR 2024) titled "Finding New Options in Biotherapeutics: Exploring Partially Morpholino-Modified Oligonucleotides as Potential RISC Users". Work contributed to the knowledge around RNA modification's effects on specific protein binding, and revealed possible unusual cases in the endonuclease activity of Argonaute-2 protein.
