About Jennifer Kay
For my graduate work, I established the cell-free metabolic engineering (CFME) group in the Jewett Lab at Northwestern University. Previous work has shown that in salt and buffer conditions designed to mimic cell cytoplasm, central metabolism can be activated to regenerate cofactors to power economical cell-free protein synthesis. It follows that central metabolism could be activated in crude lysate to regenerate cofactors for a heterologous metabolic pathway in vitro. This provides a sort of hybrid platform between purified enzyme systems and whole cell fermentation in which most soluble enzymes and some small pieces of membrane are available to an open reaction environment. Relative to enzymatic systems, it avoids protein purification costs and reduces or eliminates requirements to supplement cofactors. Advantages over traditional fermentation include direct access to the reaction without the transport barrier of the cell membrane, tolerance of many growth-toxic compounds, and the ability to more completely separate growth and production to reduce the burden of heterologous pathways on live cells. There is also potential to use this system for more HTP-friendly enzyme screening and prototyping of pathways.
