G protein-coupled receptors (GPCRs) are a large and highly varied class of transmembrane receptors that are expressed throughout many tissue and cell types. GPCRs have also been implicated in many human diseases and health conditions, and are a common target for pharmaceutical treatments. In our lab we are studying the function and expression of G protein β5, a divergent isoform of G protein β encoded by the gene GNB5, within the central nervous system. Gβ5 primarily exists within the Gβ5/ R7-RGS protein complex, and plays a role in cognitive function in humans. Our current work focuses on human and mouse phenotypes associated with GNB5 heterozygosity (GNB5 +/-), and understanding phenotypes associated with conditional Gnb5 knockout in mice as they relate to cognitive functioning. By using a combination of genetic analysis, behavioral testing, and the novel in-situ hybridization technology RNAscope, we are learning more about the expression and function of Gβ5 and the Gβ5/ R7-RGS protein complex in relation to cognition and central nervous system functioning.
