Intern
I interned in the Dynamic Biomaterials Group at the Institute for New Materials through the Cooperative International Science and Engineering Internship (CISEI). CISEI is supported by the Materials Research Laboratory (MRL) at the University of California, Santa Barbara. I had the opportunity to work with bioprinter technology, which I used in my project Melt Electrostatic Writing (MEW) of a Human Trabecular Meshwork Scaffold.
Undergraduate Researcher
Throughout the academic year, I worked with Maria Jarvis on my project, titled Time Optimization of Nanoparticle Uptake. This study compares the time-dependent uptake of different nanoparticle scaffolds within human and murine breast cancer cell lines. Particle uptake was quantified using traditional fluorescent microscopy techniques. Goals for the project were to compare the particle concentration within two different cell lines for four different particle scaffolds and find the maximum uptake within the shortest time frame. This study found that there is no significant difference in particle uptake at 15 minutes compared to 120 minutes. Results from these studies will be implemented under flow in an idealized co-culture microfluidic device.
Project Support - Laboratory
I returned to the Chemistry Group in the Research and Development sector of Abbott Diabetes Care (ADC) to continue to work on improvements for the FreeStyle® Libre Flash Glucose Monitoring System. My project was to develop a slightly tougher synthetic model of human skin that accounted for all layers of the arm tissue, this way Abbott could test prototype glucose dermal units prior to clinical trials. The end result of my project was a mixture of polyurethane and plastisol materials that could be synthesized quickly and easily. The model parameters could also be varied to mimic any part of the human body. Using previous research from the University of Michigan, I also developed a way to model the interstitial fluids in the synthetic tissue using mineral oil.
Undergraduate Researcher
I shadowed Michael Paull, a PhD candidate, on isolating antibody biomarkers in patients diagnosed with Alzheimer’s and macular degeneration. Using E. coli cells and a random library of peptides, we screened for peptides that were bound by the antibodies in the patients’ sera. Then, we separated the cells with bound peptides using magnetic-activated cell sorting (MACS). Once these cells were separated, we analyzed the cells using fluorescence-activated cell sorting (FACS). We extracted the DNA from the cells with bound peptides and sequenced it. From here, we looked for recurring peptides that were only present in disease patient sera.
Project Support - Laboratory
In this position I worked with the Research and Development sector of Abbott Diabetes Care. Currently, the R&D group is working on a continuous glucose monitoring system for diabetic patients, called FreeStyle® Libre Flash Glucose Monitoring System. This sensor stays in the arm and continuously monitors glucose levels in the blood stream. My project involved researching two improvements to Abbott’s glucose monitor, FreeStyle® Libre. The first improvement was decreasing needle size and length to incorporate a smaller sensor. Using an Instron tensile tester, I compared the force required for each needle type to penetrate a synthetic skin. The second improvement was the development of a new insertion unit. Using a high speed camera, I tracked the velocity of each inserter to monitor the consistency of the improved unit. I also designed my own fixture to be paired with the high speed camera to record the needle penetration from below the synthetic skin.
