I love working alongside ambitious people to tackle tough, impactful problems that leverage biology for good. I can be found digging into new areas (like a new lab robot or new assay), making it work, & blogging about my process. 🇨🇦 Edmonton → 🍑 Atlanta → 🦘Sydney → 🌉 San FranciscoWhile completing my PhD on bacteriophages (phages), the 'good' viruses that kill bacteria, I co-founded Phage Directory to help doctors find phage scientists willing to share their phages to rescue patients dying from antibiotic-resistant infections. Phage Directory is now the global phage community's central source of news & connection. We support thousands of phage professionals from 80+ countries, help clinicians source phages for their patients, and help organizations manage, grow & learn from their phage collections. Our 'Phage Alerts' system has helped physicians source phages for 60+ patients across more than a dozen countries, leading to at least 5 lives/limbs saved to date. In early 2022 my Phage Directory co-founder Jan and I joined the team launching Phage Australia, an Australia-wide phage therapy ecosystem. Phage Directory supported the informatics side, helping ensure the team can maximize insights it gets from its phage treatments. In this role I also dove fully back into the lab again, applying what I learned as a startup founder to designing and running the first version of Phage Australia's patient-ready phage manufacturing and QC process. We were able to set up a system that achieved a ~4-8-week turnaround per patient, 18 phage batches for 14 patients in our first year. I spent much of my time writing our SOPs, coming up with better ways of documenting and automating our process, and helping figure out how best to manage and leverage our library of hundreds of phages and unique integrated setting between lab & bedside, so we could go from 1 patient a month to 10 and beyond.As of May 2024 I've joined Paul Bollyky's lab at Stanford University to help set up a phage therapeutics pipeline to support in vivo phage delivery and cocktail development research, as well as clinical trials and compassionate phage treatments. I'm so excited to see what we can build! Postdoc, Phage Manufacturing LeadThe Westmead Institute For Medical Research Feb 2022 - Mar 2024Sydney, New South Wales, Australia- Helped establish Phage Australia's personalized, patient-ready phage diagnostics, manufacturing, and quality control system, resulting in $3.5M non-dilutive funding from New South Wales Government in July 2023.- Led production and certification of 17 phage batches approved by local and federal health authorities to be administered IV, topically and via nebulizer to 7 patients (kids and adults) across 3 hospitals through our STAMP clinical trial (one cure to date, others pending).- Implemented comprehensive processes for screening antimicrobial resistant bacterial strains, selecting active phages, manufacturing personalized batches, conducting quality checks, preparing patient-ready aliquots, delivering phages to the patient bedside, and monitoring patients during therapy.- Led the design and implementation of workflows and standard operating procedures (SOPs) for phage sourcing, biobanking, diagnostics, manufacturing, purification, quality control, and therapeutic monitoring.- Coauthored a set of regulatory documents documenting the safety of each phage batch, which gained acceptance from health district drug & ethics committees and were used to establish a standard submission process for each patient.- Acted as the main liaison between the lab and the data engineering & data science team, ensuring the informatics systems aligned with the needs of the lab and clinical settings.- Currently overseeing the transition of our phage manufacturing and quality control processes to a GMP compliant quality management system and cleanroom facility within the Sydney Cell and Gene Therapy center. -

Phd Student / ResearcherUniversity Of Alberta Sep 2012 - Jun 2018Edmonton, Alberta, Canada- I studied Campylobacter phage-host interactions, specifically how phages interact with the shape-shifting glycans of Campylobacter. Campylobacter is a difficult organism to culture and genetically manipulate, due to sub-atmospheric oxygen requirements and rapidly shifting genetic code (poly-nucleotide tracts in its genome means ~30 genes randomly toggle on and off every ~1000 divisions). To study it, we would need to make each mutation & genetic complementation multiple times in parallel due to this species' lack of clonality.- I identified a role for host oxidative stress that interfered with phage infection, suggesting previously uncharacterized metabolic requirements for the phage (Sacher et al. 2021, Sacher et al. 2020)- I studied the mechanism of antibacterial activity of a previously uncharacterized phage protein, which we named FlaGrab, with affinity for specific Campylobacter surface glycans (Javed 2015a, Javed 2015b). Through RNASeq transcriptomics I showed that FlaGrab alters metabolic gene expression upon binding, suggesting a physical mechanism triggering cells to shut down key metabolic processes. (Sacher et al. 2020)- I used RNASeq to obtain the first phage+host transcriptome of a lytic C. jejuni phage infection (Sacher et al. 2018)- I co-developed a high-throughput phenotypic screening method for identifying uncharacterized phage receptor binding proteins (Simpson et al. 2015), and adapted it to work in Campylobacter - I identified a conserved DNA modification mechanism in Campylobacter phages, explaining why native phage DNA couldn’t be PCR-amplified or sequenced (Crippen et al. 2019)- I helped elucidate the mechanism of the unexpected antimicrobial effect of cholera toxin on Campylobacter (Patry et al. 2019) -
