At UC Irvine, I have played key roles in the designing, discovery, and development of small molecules targeting protein-protein interactions of Rad51-BRCA2 and Hec1-Nek2.• Designed and developed a stereoselective route for the synthesis of a 2nd generation Rad51 inhibitor with an isoindoline scaffold, resulting in 4.8 fold improvement of IC50 in a panel of cancer cell lines.• Designed and synthesized 2nd and 3rd generations small molecule inhibitors disrupting Hec1-Nek2 interaction (INH compounds), resulting in over 10-100 fold improvement of IC50 in a panel of cancer cell lines.• Optimized the synthetic route and developed a large-scale synthesis for the 2nd and 3rd generation INH compounds to enable studies in animal models.• Guided drug designing using molecular modeling and QSAR studies for both series of inhibitors• Validation and mechanistic investigations for Rad51 inhibitors and Hec1-Nek2 inhibitors

Inhibition of the BRCA2-RAD51 interaction in breast cancer --I was interested in finding candidate drugs targeting the mechanism of DNA repair that depends on BRCA2 and an associated protein called RAD51, and rendering the cancer cells hypersensitive to radiation therapy. This would permit much smaller doses of DNA-damaging chemotherapy andradiotherapy, to lessen the side-effects associated with these treatments.
