    Chimeric Glucose Transporter Protein (GLUT 1): The purpose of this study is to understand the structure, function, and rate of sugar transport across the cell membrane of erythrocytes via a chimeric GLUT 1 transporter protein. My assignment was to develop a transient expression system for the purpose of attaining as much chimeric GLUT 1 protein as possible for further studies.Responsibilities: Site-directed mutagenesis was used for the purpose of engineering a unique restriction site in order to insert an HA -poly His tag for protein purification and Western Blot analysis. The excision of the GLUT 1 gene was from a pGEM3Z vector and ligated into a pcDNA3.1+ expression vector. This was done so as to transfect the latter into COS-7 cells, transiently express the chimeric GLUT 1 protein and purify by way of the poly-His tag. The purified protein was then to be placed into artificial membranes and measured for cell uptake and exit of sugars.
