Project 1 Overview: Colloidal stability of core-shell superparamagnetic iron oxide nanoparticles (SPIONs) as an NMR contrasting agent.• Innovated a new polymer as SPIONs coating, and designed coating grafting/exchanging route.• Investigated core-shell SPIONs chemical integrity, size, surface charge, surface coating density, colloidal stability with various instruments (GPC, FTIR, TGA/DSC, TEM, DLS/zeta potential and UV-vis spectroscopy etc.).• Purified chemicals/materials with different chromatography columns, dialysis, centrifugation and filters, and analyzed the final substance with instruments such as GPC and NMR.• Scaled-up protocols in order to better acquire various materials property data.• Developed storage methods for new molecules and biomaterials (e.g. lyophilization).• Collaborated with theoretical chemists to study the fundamental properties that impact colloidal stability of nanoparticles in the desired environments.• Analyzed data with Python, GraphPad Prism, and Microsoft Excel for publishing and presentations.Project 2 Overview: Developed highly magnetizable lipid-nanoparticles with the ability to target natural IgM-producing phagocytic B cells (NIMPAB cells) in order to develop IgM cancer vaccine.• Designed lipid coated micelles using o/w emulsion and successfully increased encapsulation efficiency of magnetic cores (3mg/ml to 5mg/ml).• Analyzed micelle stability in different solvents and measured the concentration with nanoparticle tracking analysis (NTA).• Collaborated with immunologists and developed cell assays to examine material-cell interaction, and magnetic purification efficiency. • Analyzed cell assay results with FACS and time-lapse fluorescence microscopy.• Trained and supervised one undergrad and two graduate students to develop research projects (e.g. Comsol simulation of magnetic nanoparticles’ response to external magnet with different geometries; nanoparticle formulation and stability experiments).
