I would be excited to expand these skills and apply them to novel diseases of muscle wasting and metabolic disease.In vivo experienceEstablished and characterized in vivo pre-clinical rodent models of skeletal muscle myopathies, cachexia, sarcopenia, and metabolic dysfunction. I have championed pharmacology studies that determined phamacokinetics and pharmacodynamics of low molecular weight compounds. These compounds and targets were validated in the aforementioned pre-clinical disease models for efficacy and drug safety. I established in vivo assays for evoked muscle force, voluntary running, and tumor burden. Through all of these activities, the team was able to achieve go no go decisions on targets. Ex vivo experienceI established the team's histology methods for tissue preservation, sectioning, immunohistochemistry, and automated image analysis. These techniques are routinely utilized to assess histology endpoints of fiber size, type, and regeneration. I have used a variety of biochemical assays (taqman, ELISA, etc.) to determine molecular events that explained in vivo results.

As a priority driven pharmacologist I championed in vivo studies that achieved go no-go decisions on pharmaceutical targets. By modeling metabolic dysfunction and muscle myopathy, we were able to validate targets of muscle wasting. To do so, I employed a variety of in vivo skills (compound dosing, handling, and necropsy) and was responsible to coordinate pharmacology team members to determine pharmacokinetic, pharmacodynamic, efficacy, and safety of low molecular weight compounds. I further assessed targets in these models using a variety outcomes that included insulin and glucose tolerance, body composition, muscle mass, and evoked strength. Through my ex vivo skills, I was able to generate data that helped explain efficacy results using histology, ELISA, qPCR, and multi-well plate assays. I enhanced lab productivity by training junior members to proficiently apply in-vivo and ex vivo techniques (neuromuscular function, necropsy, histology, and dosing) and by operating as the group's liaison for facility planning teams that ensured seamless function. I prepared reports and presentations were used to achieve project transitions using a variety of applications (PowerPoint, Excel, Word, GraphPad, Spotfire).

In doing so, I characterized animal models of disuse atrophy (immobilization), ApcMin+ model of cancer cachexia, sarcopenia, and diet induced metabolic dysfunction to validate targets.
