Neuman► Discovered that the RNA helicase UPF1 does not require efficient helicase activity to promote mRNA decay by building a robust workflow leveraging large computational screens, novel high-throughput in vitro assays, cellular functional assays, and NGS pipelines► Identified UPF1 mutants with predicted RNA binding alterations by performing large computational screens of over 27,000 UPF1 point mutations using python, HPC computing, Rosetta, and ViennaRNA► Measured the kinetic properties of over 30 engineered UPF1 mutants on RNA by developing 6 novel high-throughput fluorescence-based assays and increasing the throughput of existing assays ► Improved cost-effectiveness and throughput of RNA-seq by developing a novel early barcoding NGS method► Led collaborations with molecular dynamics labs and single molecule biophysics labs to measure how perturbations of UPF1 or RNA influence UPF1 dynamics and activity at the picometer scale► Mentored 6 scientists in protein engineering, nucleic acid biochemistry, and automated data analysis
