The New Target Development team at Arthrogen is focused on developing new ways to use modern molecular tools to treat inflammatory disease. Generating, purifying and characterizing novel hFIX mutants that have increased specific activity for the treatment of hemophilia B.Understanding the mechanisms of AAV-based liver mediated immune tolerance induction and applying this to human disease.

Worked with Dr. Valder Arruda and Dr. Katherine High on AAV gene therapy for hemophilia. Focusing on understanding and overcoming host immune responses. Project with Dr. Katherine High: • Investigating the immune responses to AAV capsid proteins in vitro and in vivo.Projects with Dr. Valder Aruda: • AAV mediated gene delivery using canine models of Hemophilia A to eradicate inhibitory antibodies.• Design and characterization of canine and human FIX mutants that demonstrate increased specific activity.• Investigating the use of immunosuppressive agents in non-human primate models of AAV mediated gene transfer.

Investigating the relationship between temporal regulation of antigen expression levels on the immune response using first-generation adenovirus and helper-dependent adenovirus vectors.
