I worked in the Biologics Production Facility (BPF) however, I started in the Cell Processing Facility (CPF). My duties have included the following:Produced Lentivirus used in Cellular Processing (both research and clinical use).Purified Plasmids using the Qiagen purification kits.Produced biologics using both mammalian and microbial systems.Investigated EM excursions in the BPF.Produced media and solutions for use in BPF.Perform CD4/CD8 selections on patients' apheresis product. Perform bead stimulations and lenti virus transfections on selected cells.Assist with cleaning and maintenance of incubators and BSCs.Prepared media and solutions for use in the facility.Review batch records and logbooks for accuracy and completion.

Provide expertise in transferring upstream processes from development through to manufacturing.I am responsible for New Product Introduction (NPI) for two new molecules projects into manufacturing. Investigate the use of new technologies especially single use for use in manufacturing.I am a subject matter expert on upstream operations for both mammalian and microbial systems.

My main duties are formulation and fill finish of vaccine adjuvants, and active pharmaceutical ingredients for a wide variety of external clients.At present our GMP facility is coming out of a major renovation which includes installation of a new WFI system and new equipment including an autoclave, glass washer and microfluidizer unit. I am playing an important role by assisting in the IQ/OQ plus writing the SOPs for these pieces of equipment.In addition to these roles I also receive raw materials and ship finished products to clients. Also perform all other support duties including inventory control, cleaning of facility and lab ware.

I used all my upstream experience during the start up of ZymoGenetics'​ (Bristol-Myers Squibb's) new Manufacturing Facility in Bothell, WA. My more recent responsibilities involved the start up of the new cell culture suite using my knowledge and experience with WAVE and Xcellerex bioreactor systems. I also brought my knowledge of other single use technologies including gamma irradiated assemblies, sterile welders and tubing sealers to the project. On my recommendation we purchased and use Sartorius sealers and welders. I was also author of the 4 new SOPs, for the new equipment and I assisted in the execution of the IOVs for both the WAVE and Xcellerex systems. In addition I was directly involved in the microbial upstream suite start up with a special focus on the execution of IQ/OQ of the fermentor skid in preparation for cGMP operations.Along with my colleagues, I was instrumental in the successful operation of the plant producing both microbial and mammalian projects. As a team leader and senior operator I was responsible for writing deviations and closing CAPAs. During my tenure I closed out 85 deviations in 2016, a facility record.

Duties center around cell culture operations in stainless steel and operations using single-use bioreactors. Provide technical support for different harvesting methods and technologies specifically those that make centrifugation more efficient or even unnecessary. Some of these methods include acid precipitation, flocculation, and alternating tangential flow for extended duration runs of 60 days.Also tailor and revise batch records from rough templates so they are applicable to the process at hand.

Served as production technologist in bacterial vaccines. Duties centered around fermentation of B.pertussis for Sanofi's acellular pertussis vaccine. Owned everything from producing wet seed banks to small-scale fermentation up to and including production and harvesting using centrifugation.Owned production of freeze-dried master seed bank made from the pre-master bank. In the future all pertussis vaccines made globally will be created from this master seed bank. On the development team, investigated new medium: non-animal sourced formulations for vaccine production.

Maintained mammalian cell culture stocks for use in bio-assays. Conducted bio-assays on product using MTT assay. Sampled and tested water for injection.
