• Utilized interdisciplinary background to bridge the two groups’ specialties improving communication while clarifying and realigning project short- and long-term goals resulting in two publications. • Identified failings/omissions in literature casting doubt on project viability. Adviser: Dr. Jetze TepeResearch: My research is based around proteasome manipulation. Utilizing computer modeling, I design and synthesize small molecule heterocycles as both enhancers and inhibitors of the human proteasome for use in neurological disorders and anti neoplastic activity respectively.Additionally, I explore and develop novel methodologies for the swift construction of small molecule heterocycles. In the course of this research I have:• Solved a 7 year group “paradox” by inventing an unprecedented theoretical framework for small molecule activation of the human proteasome. Also provided the first data for predicting and explaining small molecule effects on proteasome regulation in support of this framework. Results aided in securing NIH funding of ~$1,000,000 annually for the lab. Acknowledged on the group news feed January 10th, 2020.• Designed, synthesized, and tested >50 heterocyclic compounds for proteasome modulation.• Successfully modified existing commercial phenothiazine antipsychotic to the first rationally designed proteasome activity enhancer with no detectable binding of original target. • Developed new lab standard procedures in utilizing computational methods for designing proteasome modulators, and assay evaluation of synthesized compounds. • Originated 3 different projects to current multi-investigator programs in the group.• Developed the first reported, and first rationally designed, direct human proteasome activator, opening a novel area for therapeutic intervention in a host of neurodegenerative diseases.
