I'm José Pires, having recently completed my Biochemistry (BSc) and Medicine and Molecular Oncology (MSc) degrees at the University of Porto. Previously, my research at i3s delved into the intricate realm of gastric cancer extracellular vesicles glycosylation, with a specialized emphasis on truncated O-glycans. Now, transitioning to a new chapter, I am thrilled to embark on my doctoral journey at the University of Copenhagen, under the guidance of Hans Wandall. Over the upcoming three years, I will be dedicated to unraveling the complexities of carbohydrate structures within the gastrointestinal tract. Will be conducting research and pursuing my doctoral studies at University of Copenhagen (Københavns Universitet) under Hans Wandall's mentorship. Over the next three years, I'll focus on characterizing carbohydrate complexity in the gastrointestinal tract. Part of TOPGUT Marie Curie network.
Master Student
In my master's thesis project, I focused on gastric cancer, a highly lethal form of cancer with a significant global health impact. This type of cancer is often diagnosed at advanced stages and is usually asymptomatic until then, making effective treatment challenging. My research centered on abnormal glycosylation patterns, specifically the presence of a specific truncated O-glycan called sialyl-Tn (STn), which is associated with poor prognosis and tumor aggressiveness in gastric cancer. I investigated how cancer-derived extracellular vesicles (EVs) enriched in STn can influence the behavior of recipient gastric cancer cells, shedding light on their role in cancer progression. For that I took advantage of functional in vitro assays and RT-qPCR analysis. This research opened doors for further exploration of the underlying molecular mechanisms, involved in EV horizontal programming processes.

Research Trainee
This internship was a part of my First Degree in Biochemistry, in which the presence of STn both in cells and EVs of a glycoengineered cell line (MKN45 Simple-Cell), that highly expresses STn, was studied. EVs characterization was performed through western blot and their concentration and integrity though, respectively, nanoparticle tracking analysis (NTA) and transmission electron microscopy (TEM). Imuno-gold labelling TEM was performed on the EVs released by this cell line, in order to detect the presence of STn on its membrane. STn presence was also studied on gastric cancer patients tissues, through immunohistochemistry assays. All these techniques helped me to understand a little better the whole world of investigation in cancer.
