Research project: The expression of tight junction proteins in the green shore crab (Carcinus maenas) in brackish water versus seawater using RT-qPCR.• Worked on a research project in Dr. Weihrauch’s laboratory.• Designed 10 PCR primer pairs to target five tight junction genes to be tested.• Extracted RNA from the gill 8 of C. maenas using a commercial kit.• Synthesized gill 8 cDNA from the extracted RNA.• Determined the presence of the tight junction genes by performing PCR using the designed primers on the C. maenas’s gill 8 cDNA.• Confirmed the presence of tight junction genes in the gill of C.maenas by running gel electrophoresis of PCR products.• Extracted the PCR products out of the gel and purified the PCR products to be sent out for sequencing.• Determined the quality of the extracted PCR products using the Nanodrop.• Analyzed the sequenced reactions to confirm whether the right tight junction genes were presence in gill 8 of C. maenas.• Quantified the expression of each tight junction gene in the gill 8 cDNA of C. maenas under different conditions by running RT-qPCR.• Reported issues and results to the principal investigator (PI) and supervisor.• Completed documentation needed to support testing procedures.
