I’m an experienced protein engineer, project manager and leader. I’m an expert in cutting edge molecular biology techniques, translating computational protein design libraries into experimental assays, developing high throughput protocols, expressing/purifying proteins and characterizing them using various biophysical methods.As a Staff Scientist in the Mayo Lab at Caltech, I have worked on a broad array of proteins – antibodies, cellulases and methane monooxygenases – each requiring a custom expression vector and host strain (e.g. E. coli, P. pastoris). I created a streamlined process, whereby the manipulation and rapid iteration on a protein can be performed with high fidelity and with minimal hands on time. After 15 years working at the bench, you might peg me as a simple lab rat, but I haven't limited myself to just mastery of a pipette. I'm a lab manager, instrument repair guru, grant writer, human resources manager, academic-industry relationships, scientific communicator by social media and project coordinator. Broadly neutralizing antibody engineering against pandemic influenza - a collaborative project with Professor Pamela Bjorkman sponsored by DARPA; utilized yeast display to identify antibodies with increased affinity and breath to various HA subtypes from antibody libraries designed by CPDBiofuel and Natural gas-to-Liquid (GTL) fuel ProjectsIdentified cellulosic enzymes engineered by CPD for increased thermostability in a high throughput screening assay. Thermostable variants were produced in Pichia pastoris, purified to homogeneity, characterized biophysically and for their ability to maintain activity on real-world substrates.Accessory enzymes (e.g. polysaccharide mono-oxygenases) for cellulose degradation were also engineered for thermostability and shown to have a synergistic effect on the release of glucose when combined with a mix of thermostabilized cellulase enzymes. Engineered a stable, soluble and active methane mono-oxygenase under a collaborative project with Professor Amy Rosenzweig's Laboratory at Northwestern University. Responsibilities in addition to the research listed above included hiring/training/mentoring laboratory technicians, budgeting and accounting for laboratory expenses, grant writing, procurement of capital equipment, establishing a LIMS between the collaborative groups, generating timelines with deliverables, identifying solutions to problems encountered during experiments and presenting outcomes from research to collaborators/funding agencies/corporate partners. -
