    The marine diatom has great potential for application in nanotechnology, where its ordered siliceous cell wall could be used as a platform for protein expression. Bioengineering the diatom cell wall requires an in depth understanding of the pathways and factors required for targeting recombinant proteins to the cell surface. I am applying my expertise in yeast protein trafficking towards elucidating analogous mechanisms employed by the diatom, Thalassiosira pseudonana, in synthesizing its silicate cell wall and specifically targeting proteins to the cell surface. The ultimate goal of this project is to generate cost-effective, self-assembling biosensors for application in environmental monitoring, diagnostic medicine, and nanotechnology.

    The yeast vacuole is functionally analogous to the mammalian lysosome, and is best known for its function in the degradation of superfluous and damaged macromolecules. In humans, inherited mutations leading to the disruption of lysosomal function give rise to approximately 50 varieties of diseases, collectively known as lysosomal storage diseases. I used yeast as a model organism to elucidate the function of a novel vacuolar protease family member, Pff1, in order to further our understanding of vacuole and lysosome biology and dysfunction. I utilized a variety of approaches in my research including molecular, genetic, biochemical, and proteomic techniques.
