My project was titled 'Experimental validation of candidates of tissue-specific and CpG-island-mediated alternative polyadenylation in mouse'. The hypothesis was that the regulation of intragenic CpG islands activity by DNA methylation may be associated with an increase of premature polyadenylation of the host gene. To test that, I used a wide range of genetic technologies such as bisulfite sequencing, cloning, qRT-PCR, primer design and the use of specific databases like UCSC genome browser. The results of my project shed light on the role of intragenic CpG island methylation related to alternative polyadenylation as a mechanism of tissue-specific gene expression regulation. Tasks: Bioinformatics, DNA/RNA extraction, PCR, cDNA synthesis, RT-PCR, Electrophoresis, Primer design, Sanger Sequencing and Cloning (5 months at King's, 3 months home-office for writing)
