The highly collaborative, interdisciplinary, and multi-national environment at the MPI-CBG and CBSD allowed me to independently explore different topics related to biology, biophysics, imaging, image processing, modelling and simulation, and to formulate the research question of my PhD on my very own: How much to trust into the results of automated image analysis?Specifically, I worked out a spatially resolved uncertainty quantification for topological defects, which I termed "robust" defect identification. Thereby, topological defects (aka singular points) are a feature of vector and orientation fields with intriguing connections to shape, geometry and their dynamics.Starting from the mathematical definition of topological defects, and thoroughly studying its discretisation for pixelated data, I developed a measure of reliability for topological defects that are identified in noisy data, with applications from liquid crystal physics to textures to bio-medical images.Applying the newly developed methodologies to my collaborators' microscopy images of cell sheets during development allows to extract biophysical parameters that were not accessible by previous methods. This way, robust defect identification contributes to the understanding and modelling of biological tissue as active material.This position also included teaching of Saptiotemporal Modelling and Simulation to students, and supervision of course works and projects for TU Dresden students and international interns.
