Structural basis of recognition of T cell engagers that target MHC-neoantigens. - Produced and purified bispecific and full-length antibodies to be used as immuno-therapeutics for solid tumors (antibodies for MHC-p53R175H, KRASG12V, and IDH2R140Q). - Complex structures of antibodies with their MHC-neoantigen counterparts using X-ray crystallography (p53R175H, IDH2R140Q) and cryoEM (KRASG12V). - Biophysical characterization of MHC-neoantigen/antibody by SPR and thermal shift. - Determined rules of engagement for potential ‘off-the-shelf’ therapeutics. - Structural-guided affinity maturation through protein engineering. - Point of contact for CROs (molecular biology, DNA production)Regulation of the HECT E3 ligase, NEDD4L. - Determined the regulatory mechanism of NEDD4L through various biochemical in vitro and fluorescence-based binding assays. - NEDD4L inhibition studies using repurposed drugs. - Investigated the mechanism of regulation of voltage-gated sodium channels (NaVs) by NEDD4L with emphasis on protein:protein interactions, co-crystallization and in vitro assays. - Determined sites of ubiquitination of NaV1.7 by MS/MS.Trained and mentored: 1 current graduate student (Sara Nathan); 1 undergraduate student (Wendy Xia); 2 post-baccalaureate fellows (P. Mechanism of MID1, a ubiquitin RING E3 ligase. - Synthesized, purified and characterized MID1 proteins using chromatography methods, gel electrophoresis, chemiluminescent assays and dynamic light scattering. - Analyzed the effect of XLOS-observed mutations on MID1 protein structure using multi-dimensional (2D) Nuclear Magnetic Resonance (NMR). - Performed 3D NMR to solve the tertiary structure of the microtubule-associated domain of MID1. - Analyzed protein-protein interactions using numerous methods: chemical shift perturbation 2D NMR, gel filtration and native gel electrophoresis.Trained and mentored: 1 graduate, 4 undergraduate and 3 high school students.Commissioned all the cataloging and acquiring of new instruments and chemicals for the lab.
