About Kaustuv Ghosh

I have been working on understanding genetic mechanisms underlying different developmental processes. I started working on fruit flies as an undergrad in IISER Mohali, India in the lab of Dr. Lolitika Mandal. During the course of the BS-MS Dual degree, I had the opportunity to visit research centers like IISER Thiruvananthapuram and IIT Kanpur where I also got hands-on experience with other model systems like Arabidopsis, Chicken Embryo, and Mouse Embryo in the laboratories of Dr. Kalika Prasad and Dr. Jonaki Sen respectively. Later on, I decided to continue working on fly hematopoiesis and conducted my master's thesis in the lab of Dr. Lolitika Mandal. After finishing my master's degree, I moved to Barcelona where I secured the IRB DREAM fellowship to work with Dr. Marco Milan as a doctoral student. IRB Barcelona has provided me with extraordinary opportunities within the lab and outside the lab. I have been a part of the outreach activities conducted jointly by IRB Barcelona and La Pedrera Foundation. IRB Barcelona is a part of the Barcelona Institute of Science and Technology (BIST), and I had the opportunity to access BIST courses that have helped me develop skills that would stay beyond the Ph.D. Apart from the skills I have developed while conducting my research and outreach activities, I am actively looking forward to developing management and leadership skills.

Chromosomal Instabilities are genomic instabilities found in almost 90% of solid human tumors. In the Development and Growth control laboratory, we try to understand the karyotype-independent effects of Chromosomal Instability using the fruit fly as a model system. My Ph.D. work is presently focussed on unraveling genetic mechanisms involved in controlling the tissue dynamics in the tumor model. We have recently shown how sub-lethal caspase levels activated as a consequence of chromosomal instability play a role in tissue invasiveness.

Drosophila hematopoiesis takes place through two distinct waves of hematopoiesis. The second wave or the definitive wave of hematopoiesis gives rise to mature hemocytes that populate the organism during the larval and the adult stages. These hemocytes emerge from hematopoietic progenitors housed in an organ called the Lymph gland. The hematopoietic progenitors have recently been shown to require Fatty-Acid β Oxidation (FAO) to stay in quiescence and later, to differentiate. Interestingly, we found that these progenitors are initially proliferative and then turn quiescent. We asked whether the early and late progenitors have distinct metabolic landscapes, which in turn manifests as two different cellular states in these cells. We found that early progenitors exhibit extensive glycolysis in stark contrast to the late progenitors which rely majorly on FAO. Our results indicate that the role of an extrinsic signal is essential for regulating glycolysis in the early stage to help these progenitors divide and subside as these progenitors become quiescent. In conclusion, the work conducted describes a metabolic basis of how the organ generates the required stem cell/ progenitor pool in the early stages in collaboration with cellular signaling and later shuts this metabolic program to differentiate and become functional. The long-term goal of this project would be to find out how cellular signaling affects the production of ROS and to specifically investigate whether it abrogates mitochondrial physiology or it enhances the activity of cellular ROS scavengers. It would be also interesting to investigate how this signaling is affected in various pathophysiological conditions to modulate hematopoiesis. During infection, progenitors have to differentiate precociously, it would be interesting to further note, whether a metabolic dynamism is required to elicit immune responses.
