o Ultrapure DNA preparation- I developed and optimized proprietary techniques for ultra-pure genomic DNA purification from diverse organisms across kingdoms. The DNA is used for whole genome NGS using PacBio, Illumina, and 10X Genomics

o Invented a novel biotherapeutic for smoking cessation• Hapten carrier design and synthesis-used in silico techniques to design a novel trimeric coiled-coil hapten carrier for coupling nicotine (and other small molecules). Synthesized the vaccine in vitro and characterized immune response and function. Received a US and European patent for this technology.• Vaccine characterization- B- and T-cell assays measured vaccine efficacy in mice. The vaccine produced a long-lived high titer IgG response and functioned to inhibit nicotine from crossing the blood-brain barrier.

o P.I. of the Immune scFv Laboratory. Secured $1.2M in funding for my own program and was part of a collaborative team that acquired about $4M in fundingo Engineered and produced novel recombinant mouse and human scFv antibodies for diagnostic and detection assays.• Yeast surface display and FACS were used to engineer and select single chain antibody fragments (scFv) for use in many human health diagnostic and detection assays.• Functionally tested scFvs following expression in E. coli or yeast. Antibodies were used in ELISA and microarray assays and performed as well as IgGs. Immunized mice were discovered to be a superior source of antibody libraries following antigen-specific FACS sorting of lymphocytes.o Designed and constructed two novel sensor proteins• Engineered novel TNT and RDX detection platforms- TNT and RDX-specific scFv antibodies were fused to GFP and coupled with soluble luciferase for BRET assays to detect ultra-low levels of explosives in the environment.• Engineered a novel uranium detection protein- I developed a novel uranium sensor protein comprised of a circularly permutated calmodulin-GFP isoform for detection of uranium ions in the environment.

o Optimized recombinant human scFv antibody expression in bacteria and yeast. • scFv antibodies were selected by antigen-specific FACS sorting of yeast-Selected genes were amplified by PCR and ligated into E. coli, S. cerevisiae, and P. pastoris soluble expression plasmids for production.• Optimized scFv antibody production-secreted soluble scFv antibodies were processed and purified from media (yeast) or the periplasm (E. coli). Functional activity was tested using surface plasmon resonance spectroscopy.

o Human recombinant IgG expression in plants• Patented a novel promotor to drive recombinant IgG gene expression in plants- Received a patent on this technology in 2005• Developed of a novel aqueous two-phase partitioning method for pigment removal- I developed a novel aqueous two-phase partitioning technique to remove plant pigments (primarily chlorophyll and some flavonoids) from crude extracts facilitating downstream recombinant IgG purification.

•Biochemically and biophysically characterized a novel peripheral membrane calmodulin binding protein in elongating pollen tubes of Petunia hybrida (this protein is also present in other Solanaceous species including potato, tomato, and tobacco).•Characterized the localization of this protein to novel organelles in elongating pollen tubes.•Characterized tissue specific expression in whole plants.•Determined gene copy number, sequence, and structure.•Compared sequence to database proteins and used bioinformatic information to define function.
