Molecular Cell Biology:• Transformed and selected bacteria, and purified plasmid containing shRNA or gene construct• Packaged plasmid into viral vector for transducing leukemic stem cell (LSC) models• Transduced LSC models with CRISPR construct and isolated and selected single colonies• Maintained several cell culture types including suspension, adhering, and semi-solid media• Performed western blots and qPCRs for analysing changes in expression of genes of interest• Prepared cytospin slides with fluorescent antibodies and conducted confocal microscopy• Utilised flow cytometry for performing cell cycle, apoptosis and cell sortingMouse work:• Handled and monitored C57BL/6 and NOD/SCID mice weekly• Humanely euthanised and harvested tissues to process, store and perform experiments on• Experience with IV, IP and blood collection of mice
