    Vascular Medicine InstituteIndependent Research ProjectWorked on a project focused on the lipid binding and peroxidase activity of Cytoglobin(Cygb). Cygb is a heme-containing protein that has shown protective effects in brain injury models. The project tested the hypothesis that Cygb is a peroxidase activated by lipids that catalyzes the formation of lipid mediators. To determine the lipid binding site, we generated mutations of Cygb around the locations identified in preliminary studies determined the peroxidase activity of wild-type and mutant Cygb in vitro in the presence of physiologically relevant lipids. Cygb mutants with gain/loss of function can be applied to study the role of Cygb in cell systems.Team Development-Collaborated with the research team on a weekly basis to communicate our results and determine new actions as necessaryLab Technique-Recorded the lipid binding specificity of various mutated Cygb using photospectroscopy.-Collected protein via Affinity Chromatography.-Diluted lipids and proteins within the magnitude of nano molar range.-Followed strict protocol for handling hazardous material.
