Research Focus: Generating human primary nociceptive (pain sensing) neurons using directed differentiation, and understanding transcription factor codes in directed differentiation of specific neuronal subtypes • Developed a novel transgenic technique to generate functional somatosensory neurons (neurons that sense pain, temperature, itch, touch, and muscle position) from multiple tissue sources including mouse and human fibroblasts, induced pluripotent stem cells, and embryonic stem cells. • Implementing single cell RNAseq to validate discrete neuronal subtypes in both distinct neuronal populations and in induced neuronal populations. • Genetically engineering induced somatosensory neurons using CRISPR/Cas9 to generate an assay for high throughput pharmacological screening. • Established a large screen to transgenically generate a wide array of functional neuronal subtypes in vitro using combinations of transcription factors. This screen has generated dozens of novel in vitro neuronal subtypes that are currently being characterized and validated for use as human neuronal models. • Implemented a viral technique developed in the Baldwin lab to trace single olfactory neurons throughout their projection along the entire length of the mouse brain. Single axon traces in different genetic backgrounds tests for the effect of sensory input on the axonal connectome.
