A 12-month internship in the Biosciences Division at the Eugene site doing product development and research activities for cell-based imaging tools and bioassays of novel fluorogenic detection reagent candidates for drug discovery and basic sciences research. • Testing and structure- activity relationship characterization of novel, activity-dependent caspase indicator fluorescent dye tools.• Duties include testing customer and product requirements to set specifications on dye candidates in research test case scenarios. • Workup and assay development of cell stress, inflammation, and programmed cell death model systems. • Functional characterization of dye activity and quantification of key performance indicators in cell signal, percent responders and selective inhibition metrics. •Mastered basic and advanced quantitative technologies on the EVOS and CellInsight imaging systems to assign pass and fail criteria to dye candidates and results.• Regular report outs and collaborative discussions with external team of biologists and chemists for peer-informed design of experiments and test conditions.• Maintain cell culture of human and transformed cell lines in a biosafety Level 2 lab. Dr. Matthew Posewitz Research Group - https://chemistry.mines.edu/project/matthew-posewitz/• Investigated water sustainability and land availability challenges in the food sector by studying the diverse portfolio of protein, lipid, and carbohydrate bioenergy sources of 3 distinct strains of the marine alga Picochlorum celeri.• Served as the primary point of contact for all HPLC sample analyses for the lab group, in addition to supporting general routine maintenance of the instrument. • Determined impact of growth conditions on chlorophyll content for 3 algal strains through HPLC operation, data organization in excel, and analysis for pigment quantification of strains under nutrient stress growth regimes. • Refined sample preparation methods through iterative testing of the lab’s standard procedure to identify the most efficient conditions for pigment analysis and standard curve generation.• Identified a less hazardous solvent (acetone vs petroleum ether) to use in UV-Vis studies to improve lab safety through extinction coefficient determination of the pigment canthaxanthin.• Ensured algal culture health through daily care including inoculation, regular monitoring, sterile cleaning, nutrient and chemical preparation, and autoclaving for flask and media equipment sterilization.• Presented research findings at an undergraduate research symposium and during bi-annual presentations to enhance communication skills with individuals of varying levels of foundational and scholarly knowledge.
