The Huang Lab is a microbiology laboratory focused on understanding how bacterial cells grow and divide and how these basic factors impact basic science, biotechnology, and medicine. My main project consists of characterizing and quantifying bacterial evolution in the intestine utilizing a library of DNA barcodes that I constructed to track Bacteroides thetaiotaomicron both in vivo and in vitro. Additionally, I used confocal microscopy to live-image a library of 300+ gut commensals to quantify single-cell growth dynamics.

The Rosenblum Lab is a basic immunology laboratory. The goal of our research is to identify the cellular and molecular mechanisms responsible for the establishment and maintenance of immune tolerance to antigens expressed in tissues. In addition, we aim to identify the mechanisms responsible for T cell-mediated autoimmune diseases. Our research utilizes animal models of inflammation and autoimmunity as well as human tissue from patients with malignancy and autoimmune/inflammatory diseases.

As a member of the enteroendocrine cell (EEC) program team, I evaluate potentially bioactive peptides in mouse models of type 2 diabetes. I perform a wide variety of in vivo and ex vivo procedures, including animal grouping, tail vein injection, bleeding, weekly measurements of blood glucose and body weight, glucose tolerance tests, animal sacrifice, ELISA analysis, and DNA amplification and purification. I also analyze and present data in bi-weekly meetings. Identified one novel peptide with confirmed glucoregulatory activity.

Under the general direction of the LA III, lab supervisors, and using standard operating protocols (SOPs), manuals, Environmental Health & Safety (EH&S) guidelines, reference books, and scientific literature as principal sources of technical information, I independently went to any one of 20 labs on campus to wash glass- and plastic-ware by hand or by machine. Prepared glassware for sterilization and sterilize using autoclave, wash pipets, and restock shelves, drawers and cupboards with clean, dry glassware. Kept sterile glassware and equipment separated from non-sterile stocks. Maintained order and cleanliness of glassware washing areas.

I purchased, databased, and tested products with possibly harmful chemicals to help identify those that may be in violation of Prop 65. Assisted with general CEH organizational support as necessary, including occasionally pitching in on other program work. My work aided in a landmark legal agreement with companies like Playtex, West Elm, and dozens of other companies to end the use of harmful chemical flame retardants in California.

I assisted in the maintenance and setup of experiments performed in the undergraduate organic chemistry lab classes. I also helped maintain the supplies and equipment in the organic chemistry storeroom, and issued lab equipment and chemicals to lab students during classes. I performed general cleaning and restocking of the organic lab rooms and general maintenance of the instruments used in the organic lab rooms.

Most of our research focuses on native fishes in their natural habitats, and involves sampling fishes and the physical habitat via standard methods in stream and estuarine ecology. I assisted a PhD student carry out a population and species survey using electrofishing equipment and pit tagged fish in the parr stage. I performed weekly measurements of stream volume and fish body weight and length. Analyzed samples of stream leaf litter and performed insect population estimates. I also analyzed the data and presented it at a symposium and national conference.

I feed, cleaned, and cared for 60 species of animals (reptiles, mammals, amphibians, etc.), administered vet care, and performed miscellaneous husbandry duties. I also worked with and taught museum visitors of all ages in the Animal Discovery Room. I mentored and supervised students (ages 15-17) in the TEAMS Youth Intern Program, and attended monthly staff meetings.
