    My PhD is funded by the Medical Research Council Doctoral Training Partnership (MRC DTP) and involves engineering liposomal nanomedicines capable of selective and targeted drug release for advanced prostate cancer. My project is joined between the Androgen Signalling and Prostate Cancer Group (led by Professor Charlotte Bevan) and the Membrane Biophysics Group (led by Professor Oscar Ces). During the project, I have:•Gained extensive experience in mammalian cell culture, 3D cell culture, cell viability assays, RT-qPCR, and western blotting •Gained expertise in culturing patient biopsy samples and preparing samples for immunohistochemistry (IHC) •Successfully planned and executed an in vivo study evaluating the biodistribution and toxicology of our liposomal formulations •Developed and optimised a robust, high-performance liquid chromatography (HPLC) pipeline to quantify drug loading into liposomes

    I was a member of the Androgen Signalling and Prostate Cancer group led by Professor Charlotte Bevan. My project involved exploring the mechanism of action of CT7001 (ICEC0942), an orally available ATP-competitive cyclin-dependent kinase 7 (CDK7) inhibitor developed at Imperial College London. During the project, I was able to successfully:• Explore the side-effects of orally administered CT7001 on the small intestine and liver using immunohistochemistry (IHC) staining• Employ site-directed mutagenesis and luciferase-based reporter assays to investigate the importance of post-translational modification of the androgen receptor to the action of CT7001• Evaluate the efficacy of CT7001 against constitutively active androgen receptor mRNA splice variants

    I was a visiting scholar at The University of California, San Francisco (UCSF), supervised by Dr Lisa Chesner and Professor Felix Feng. My project involved investigating the regulation of Major Histocompatibility Complex (MHC) class I and II in prostate cancer. Overall, the project aimed to improve the efficacy of immunotherapies, such as immune checkpoint blockade, which has shown limited clinical benefit in immune-cold tumours such as prostate cancer. During the project I successfully planned and performed a CRISPRi flow cytometry screen to identify regulators of MHC.

During the project, I was able to successfully:• Utilised P1 phage transduction to generate in-frame single-gene knockout mutations in several essential TCA cycle genes•Developed a robust and rigorous method for conformation and screening of gene-knockouts• Learned advanced microbiology techniques used to investigate DNA methylation stability in the absence of gene regulation•Verified the activity of DNA methyltransferases (DNMTs)
