Lead their work and my own on projects including: 1) improving gene editing using lentivirus, AAV, mRNA transfection, and site specific nucleases (CRISPR/Cas, TALENs, ZFNs, mTALs, and Homing Endonucleases); 2) developing ddPCR-based high-throughput analysis of gene editing (deletions & HR); 3) creating algorithms for selecting optimal targets for genome editing and homing endonuclease re-specification for those sites; and 4) development of a clinically relevant technology to improve mRNA-based gene editing and personalized medicine. For the latter, created white papers, worked directly with legal team to prepare patent applications, and managed external collaborations and sponsored research agreements. Lead the creation of primary tissue culture SOPs. Organized the lab by assembling our strains, constructs, and orders into an electronic database and creating guidelines for its use; by creating best practices for use of a program-wide electronic lab notebook; and by implementing and training our program to use an online project organization tool. Created and maintain a server for data analysis tools for the lab and am the ‘go-to’ person for technical needs.

Developed a new high throughput method for characterizing the specificities of site-specific nucleases (for genome editing in basic sciences, biotechnology, and clinical medicine). Designed the method to tie into our existing flow cytometry-based characterization and directed evolution platform, and created custom software to search for patterns in the resulting next generation sequencing data.
